Been meaning to post something like this for a while, but here goes -- and feel free to join in! Here is the list of my current "must read" webcomic links:
Ctrl+Alt+Del (http://www.ctrlaltdel-online.com/) - Has a very good record for spot-on video game jokes, but seriously lacks in the ongoing story department (crazy video game player, yawn).
Penny Arcade (http://www.penny-arcade.com/) - Still awesome. My only complaint is that their front page posts are less informative and interesting as they used to be.
Real Life (http://www.reallifecomics.com/) - Hit and miss, not crazy about the graphic style, but there are some funny jokes and it goes down smooth.
VG Cats (http://www.vgcats.com/) - Awesomely funny, twisted comics about games, starring cats. Very infrequent updates, tho. Check out the 300 parody strip, definitely my fave.
Order of the Stick (http://www.giantitp.com/comics/oots0484.html) - A serial D&D comic strip. Often hilarious, with multiple jokes per strip, and plenty of geeky in-jokes. On the downside, the last 100 strips or so seem like a boring drawn-out battle that makes me yearn for back when they dungeon crawled. Also, the fans of this strip treat it like it's the holy grail or something, and hang on every word, which irks me.
Perry's Bible Fellowship (http://pbfcomics.com/) - Crazy variety on art styles, twisted beyond belief, and drop-dead funny stuff. Updates once a blue moon, however.
XKCD (http://xkcd.com/) - A newer fave of mine, this stick figure strip is as nerdy as they come, and that's the best part. Some of the jokes are far above my head, with mathematical/scientific/logic terms, but I tend to get most of them, and he's just great with the jokes. Also, make sure to mouse over to read the alt text, which continues the strip somehow.
Dark Legacy Comics (http://darklegacycomics.com/) - A World of Warcraft strip. Infrequently updated, and mostly pertains to the game, but it's pretty funny even if you don't play. Well, probably not AS funny.
Crimson Dark (http://www.davidcsimon.com/crimsondark/index.php) - An ongoing scifi serial strip, done with Poser. The creator is an admitted fan of Firefly, and the strip is a sort-of homage to the show -- a group of misfits on a transport vessel, etc, with secrets, etc etc. But it's light on its feet with the jokes, the story keeps interest, and it's not half bad to look at.
The Non-Adventures of Wonderella (http://www.graphicsmash.com/comics/wonderella.php) - Hands-down, my favorite webcomic. Updated Saturdays, this strip continually floors me with its weird humor and situations about a superheroine who couldn't care less about her job, and talks like... well, like someone who couldn't care less about English, either. You have to love her pluck, tho!
Lio (http://www.gocomics.com/lio/) - The only syndicated strip I read online, Lio just appeals to me with its silent (no dialogue) love of horror, scifi, technology, and all sorts of things boys loved to dream about when they were kids. Kind of like Calvin and Hobbes, just more enthusiastic about the mad scientist stuff.
Hammer of Grammar (http://meghanohara.org/wordpress/) - Another World of Warcraft strip, EVERYONE should give this a look. The author is terrific at nailing jokes, even if you don't play the game.
Basic Instructions (http://www.basicinstructions.net/) - A strip drawn by a stand-up comedian on "How To Do" most everything. Very hilarious, and the "How To Fake A Smile" had me laughing crazy-like for a half hour.

PvP - Stopped being funny, stopped being about games, started being a lot about making money with advertising and how important Scott Kurtz is.
Shortpacked - Very funny when it's about toys, movies or games, but the normal story isn't nearly as good.
